MppTAC3 missing experimental subject YFP fusion (EFp::MppTAC3-YFP) was expressed and imaged in isolated chloroplasts against DAPI-stained nucleoids. MppTAC3 was strictly co-localized with cp nucleoids in M. polymorpha. Proposed as a eukaryotic DNA-packaging component replacing prokaryotic HLP during land plant evolution.
MpSWIB missing experimental subject YFP fusion (EFp::MpSWIB-YFP) was expressed and imaged by confocal microscopy with chlorophyll autofluorescence. MpSWIB localized mainly to nucleus and cytosol, not chloroplasts, indicating it is likely a nuclear factor rather than a cp nucleoid component.
Mp3g01640 MpWHIRLY experimental subject YFP fusion (EFp::MpWhirly-YFP) was expressed and imaged in isolated chloroplasts with DAPI-stained cp DNA. MpWhirly co-localized with chloroplast nucleoids, with signal also in the extra-nucleoid region. Studied as a candidate eukaryotic-derived cp nucleoid component in M. polymorpha.
