The MpMME1 malic enzyme homolog was amplified from Marchantia cDNA, fused to a fluorescent reporter and expressed in Marchantia to test targeting. Its mitochondrial targeting peptide normally directs it to mitochondria; when the positively charged residues of the mTP were substituted with serines the construct instead predominantly targeted plastids, showing that charge divergence in the targeting sequence determines organellar specificity.
