MpARF1 Transcription factor, similarity to Arabidopsis activator ARFs. Published

Associated Literature (AI-based literature mining, Beta-version)

Transcription factor, similarity to Arabidopsis activator ARFs.; The M. polymorpha genome encodes three ARFs (MpARF1, MpARF2, and MpARF3). The three MpARFs have a structure common to typical ARFs, containing a B3-like DNA binding domain (DBD) and a protein-protein interaction motif, called domains III and IV (Figs 1A and S2). Between the DBD and domain III, MpARF1 harbors a glutamine-rich region, which is known as a feature of activator ARF members. Phylogenetic analysis showed that MpARF1 belonged to the clade A, which represents activator ARF proteins such as ARF5/MONOPTEROS (MP) of Arabidopsis. (from nomenclature)

The single A-class ARF; Mparf1-kd shows MpARF1-mediated auxin responses antagonize both auxin production and MpARF2 expression in different domains.

Activating A-class auxin response factor. This study showed MpARF1 shares the conserved arginine-dependent degradation mechanism with MpARF2, but stabilizing MpARF1 had little effect on development or auxin response, so its proteolysis is functionally dispensable.
Rienstra, J., et al. (2025) · Proceeding of the National Academy of Sciences USA  research experimental subject
Functionally studied as the sole A-class positive effector of auxin transcription: alanine substitutions (R194, P197, R199) and B3-domain swaps were assayed for complementation of the Mparf1-4 null. RNA-seq showed Tak-1 responded to IAA (112 DEGs) while Mparf1 showed no auxin response, defining R194/P197/R199 as essential for its DNA-binding function.

Auxin response factor; in N. benthamiana agroinfiltration assays MpARF1 induces the proMpACL5::LUC reporter, linking auxin signaling to the thermospermine pathway.
de Roij M, Hernandez Garcia J, Das S, Borst JW, Weijers D. (2025) · Nature plants  research experimental subject
Experimentally studied. The A-class ARF MpARF1 protein is likewise shown to be unstable, and degradation-motif swap experiments confirm it shares the conserved degradation determinants characterized for MpARF2.
